摘    要:以猕猴桃(Actinidiaspp.)幼叶为材料提取基因组DNA,从64个引物组合中选取12对引物进行雌、雄集群DNA样品的扩增片段长度多态性(Amplified fragment length polymorphism,AFLP)扩增,再从中选取4对引物进行6对雌、雄DNA样品的AFLP扩增.4对引物共扩增出74个AFLP标记,其中多态性标记为58,多态性标记百分比为78.4%.遗传相似性分析表明,同一品种雌雄样品之间的AFLP电泳图谱表现出一定的差异,但小于不同品种间的差异,而品种间的差异又小于不同种间的差异.同时AFLP多态性初步检测出猕猴桃不同性别在DNA水平上的差异.

Abstract:Genomic DNA was extracted from kiwifruit leaves and.AFLP analysis of male and female plants of 6 cultivars or species was preliminarily conducted.Four primer pairs were selected for AFLP amplification and 74 reliable AFLP amplificates were generated,of which 58 were polymorphic,revealing 78.4% of polymorphic markers in genus.Genetic similarity was analyzed using reliable AFLP markers through NTSYSpc(Version 2.10e) software.The dendrogram was constructed based on UPGMA clustering analysis for establishing an evolution tree by using SAHN software.The DNA polymorphism of female and male in Actinidia were screened and different bands of AFLP amplificates were present in the electrophoretogram.The experimental results showed that differences between female and male of the same cultivar were less than those between different cultivars,and that differences between different cultivars were less than those between different species.Differences between female and male of the same cultivar or species suggested that AFLP could be applied in kiwifruit genetic studies related to sex.By means of AFLP analysis,genetic diversity and species biology of kiwifruit could also be studied at molecular level.
