摘    要:将菜豆细菌性萎蔫病菌(Curtobacterium flaccumfaciens pv. flaccumfaciens)16S rDNA基因进行克隆测序,并依据此序列设计菜豆细菌性萎蔫病菌的实时荧光PCR引物和特异性探针,对所收集的样品进行了检测. 结果表明,该检测方法具有快速、特异、灵敏、重复性好等优点,只有菜豆细菌性萎蔫病菌产生荧光,其他细菌没有荧光产生. 检测的灵敏度为105 CFU/mL的细菌悬浮液,相对灵敏度为106 CFU/mL.

Abstract:The 16S rDNA gene of Curtobacterium flaccumfaciens pv.flaccumfaciens was cloned and sequenced, and it was detected by real-time fluorescent PCR. A subgroup-specific probe was designed based on its sequence, which was used to detect 10 bacteria. The results showed that no signal could be detected from the other bacteria except Curtobacterium flaccumfaciens pv. flaccumfaciens with the relative sensitivity was 10~6 CFU/mL and that of the suspension of pure culture was 10~5 CFU/mL. The described method has the advantage of high speed, sensitivity, specificity and stable reproduction.
