肥育猪背最长肌NF-κB,IκBα和p94蛋白表达量与其剪切力的相关性研究

摘    要:研究饲粮不同蛋白水平对猪骨骼肌中NF-κB、IκBα和p94蛋白表达量的影响,并探讨其与剪切力的相关性。试验选用90头约50kg的杜洛克×长白×大白三元杂交猪(公母各半,公猪去势),随机分为3个处理组,每个处理组3个重复,每个重复10头,分别饲喂蛋白质水平为12%、16%和20%的饲粮,各饲粮能量及氨基酸模式相同,饲养58d后屠宰取样,用蛋白质免疫印迹法测定猪背最长肌中NF-κB、IκBα和p94在骨骼肌中的蛋白含量。结果表明:饲粮蛋白水平对猪骨骼肌中NF-κB蛋白含量影响不显著(p>0.05),但对IκBα和p94蛋白含量和肌肉剪切力影响显著(p<0.05),且随着饲粮蛋白水平升高,肌肉IκBα蛋白含量呈升高趋势,p94和NF-κB蛋白含量呈降低趋势,肌肉剪切力呈升高趋势。肌肉IκBα蛋白含量与剪切力呈正相关,p94蛋白含量与剪切力呈负相关,NF-κB蛋白含量与剪切力不相关。

Abstract:This study aimed to examine the effect of dietary protein levels on NF-κB,IκBα and p94 protein expression and muscle shear force in swine pig skeletal muscle.Ninety crossed-bred(Duroc×Landrace×Yorkshine) pigs with an average weight of 50 kg,were randomly assigned to three dietary treatments with three replicates and ten pigs in each pen,and fed with diets of three protein levels(12%,16% and 20%) respectively.The energy level and amino acid pattern in the three diets were kept the same.The whole feeding trial lasted for 58 days.Then Western Blotting was used to determine the proteomics relative expression of NF-κB,IκBα and p94 in skeletal muscle.The results showed that different levels of dietary protein had no effects on the NF-κB(p>0.05),but had effects on the IκBα and p94 proteomics relative expression and the muscle shear force in skeletal muscle(p<0.05).With the increase of diet protein level,the relative expression of IκBα and the shear force in longissimus muscle increased,the relative expression of NF-κB and p94 decreased.The relative expression of IκBα correlated with muscle shear force positively,but p94 correlated with muscle shear force negatively,while NF-κB did not correlated with muscle shear force.
