摘 要: | 脯氨酸的积累与植物对干旱和盐胁迫的抗性有关.P5CS基因编码一个双功能酶,具有谷氨酸激酶和谷氨酸-γ-半醛脱氨酶活性,催化脯氨酸合成过程中的前2步反应.它是脯氨酸合成的限制因子.用生物素标记的荧光原位杂交技术,以蛾豆(Vigna aconitifolia)中克隆到的P5CS基因作探针对水稻进行物理作图.该探针定位于水稻第9染色体的短臂近端点处,信号点距着丝粒的相对距离为(51.79±1.24)%.P5CS基因广泛存在于单子叶和双子叶植物中.

Abstract: | Proline (Pro) accumulation has been associated with tolerance to drought and salinity stresses inplants.△'-pyrroline-5-carboxylate synthetase (P5CS) gene encoded a bifunctional enzyme with both g-glutamylkinase and glutamic-g-semialdehyde dehydrogenase activities that catalyzes the first two steps in proline biosyn-thesis.It was rate limiting for Pro synthesis.Using a biotin-labeled fluorescent in situ hybridization (FISH)technique,P5CS cDNA clone from mothbean (Vigna aconitifolia ) as a probe was approximately located onto thelong arm of rice chromosome 9.The percentage distance (from the hybridization sites to centromeres) was(51.79±1.24)%.P5CS is widely present in monocot and dicot. |
