玉米EST,GOT,CAT等位酶基因的表达特异性研究

摘    要:采用平板淀粉凝胶分析技术,对5种不同遗传背景、7种不同取样处理的玉米材料进行酯酶(EST)、谷草转氨酶(GOT)、过氧化氢酶(CAT)9个位点的等位基因表达特异性的研究。结果表明,在不同遗传材料之间EST差异明显;同一单株不同组织样品之间EST差异不大;3个GOT,2个CAT位点的基因在不同的组织样品之间存在明显的表达时序性,基因表达受到发育阶段的控制;CAT2基因的表达受光照诱导,GOT1基因的表达受低温刺激诱导,以上5个位点的等位基因在各种遗传材料之间差异不大;正常胞质与不育胞质对等位酶基因的表达在苗期没有影响。

Abstract:The EST GOT and CAT allozyme of maize were analyzed by hrizontal starch gel eletrophorsis.The samples for seven treatments were gathered among five different genetic background materials of maize. The results indicated that the allpzyme pattern varied greatly between the different samples, and some tissues had a characteristic and distinguishing complement.Ontogenetic variations on the expression of allozyme gene were observed for coleoptil,sheath,green leaves, low temperature, mesocoty, roots, colluse. The EST pattern was changed greatly under different materials, but it was slightly altered under different tissues and treatments. The GOT and CAT allozyme profiles have more different expressions among the different tissues and treatments than among the different materials.No influence for above three kinds of allozyme gem's expression was found in cytoplasm of male sterile maize. The temporality and spatiality of allozyme gene's expression not only influenced by genetic factors,but also responsive to exogenously applied environmental signals (physical and chemical).The light induced gene of CAT2 to expression and the low temperature led gene of GOT1 to expression.
