电场诱导融合再生两个柑桔种间体细胞杂种

摘    要:通过电场诱导融合方法分别获得了续荷尔脐橙与粗柠檬、纽荷尔脐橙与夔柑两个组合的体细胞杂种。纽荷尔脐橙+粗柠檬级合获得了45棵再生植株。对其中的30棵株进行染色体检查,结果表明,全为四部体,2n=4x=36。过氧化物同工酶及RAPD分析表明再生植株为体细胞杂种。续荷尔脐橙+夔柑组合获得了25株再生植株。染色体检查结果表明,全为四倍体,2n=4x=36。RAPD分析表明再生植株为体细胞杂种。

Abstract:Protoplasts of the embryogenic callus of Newhall navel orange (Citrus sinensis) were electro-fused with mesophyll protoplast s of Rough Lemon (C. jambhiri) and Kuigan tangelo (C. reticulata × C. grandisi ) respectively. 45 and 25 plants regenerated from these 2 combinations respectively. Chromosome cou nts of 30 plants of the combination of Newhall navel orange +Rough lemon, and 25 of Newhall + Kuigan showed that all of them were tetraploids (2n=4x=36). Peroxi dase isozyme and RAPD analysis verified that the tetraploid plants of Newhall + Rough lemon were somatic hybrids. RAPD analysis with primer OPA14 and OPA08 toge ther verified that all of the tetraploids derived from Newhall+ Kuigan were also somatic hybrid plants.
