唐菖蒲花茎愈伤植株再生途径的优化

摘    要:研究了外植体不同生长阶段、激素种类和浓度对2个唐菖蒲品种愈伤组织诱导及不定芽再生的影响。结果表明,以Rose Supreme 5叶期和Advanced Red 7叶期的花茎为外植体较好,2个品种直接诱导愈伤组织及不定芽再生的最佳培养基为MS BA 2 mg/L IBA 0.4 mg/L,愈伤诱导率分别达到80.50%和87.78%,愈伤全部再生不定芽,平均再生芽数分别为14.2和13.0,培养周期仅为45 d左右。在生根培养基MS IBA0.5 mg/L上生根率分别达86.67%和92.44%,单苗根数为16.1和22.1,根势好,结球率均达90%以上。

Abstract:The effects of period of explants and growth regulator on the callus induction and adventitious shoot formation were investigated.The results showed that scapes within five-leaf period of Rose Supreme and seven-leaf period of Advanced Red were ideal explants.MS BA 2 mg/L IBA 0.4 mg/L was the best medium for callus induction and adventitious shoot formation,in which callus percentage of Rose Supreme and Advanced Red separately was 80.50% and 87.78%,shooting percentage was the same with that,and average adventitious shoots per explants were 14.2 and 13.0 respectively,culture period was just about 45 d.When the medium for rooting was MS IBA 0.5 mg/L,the rate of rooting reached 86.67% and 92.44%,the number of roots per plantlet were 16.1 and 22.1,and the roots grew strongly and the rate of corm formation was above 90%.
