SARS冠状病毒刺突蛋白S144-643的表达及免疫原性分析

摘    要:将SARS冠状病毒(SARS-CoV)表面的主要结构蛋白刺突蛋白S144-643的基因片段(nt430-nt1929)克隆入表达质粒pET-28b中,并在大肠杆菌中表达。S144-643蛋白分子质量为55 ku左右,以包涵体形式存在于细菌中。ELISA及Western blot证明,S144-643可以与SARS康复病人的阳性血清发生特异性反应,免疫新西兰大白兔可以诱发产生较高水平的特异性抗体,因而证明原核表达的S144-643具有良好的免疫原性。

Abstract:The gene fragment of S144-643 was subcloned into pET-28b and expressed the protein in E.coli.The protein existed mainly in bacterial inclusion bodies.The S144-643 was purified from bacterial inclusion bodies,refolded with the refolding buffer containing reduced glutathione and oxidized glutathione.SDS-PAGE demonstrated the protein with the molecular mass of about 55 ku.The immunogenicity of S144-643 was demonstrated by ELISA and Western blot assay,while the recombinant protein was specifically recognized by the sera from convalescent SARS patients.Furthermore,S144-643 was able to induce high titer of specific antibodies in immunized New Zealand rabbits.The S144-643 protein was expressed in E.coli and was immunologically active.
