新城疫病毒F-HN基因DNA疫苗的免疫效果评价

摘    要:新城疫是由新城疫病毒引起的一种高度接触性传染病,严重危害全球养禽业.DNA疫苗因其独特的安全、有效等特点将会是解决传统疫苗不足的新途径.为了评价NDV保护性抗原F、HN融合基因的效果,以PBS作为空白对照,将pVAX Ⅰ、pVAX Ⅰ-F、pVAX Ⅰ-HN和pVAXⅠ-F-HN四种质粒肌肉注射2周龄雏鸡,一免后二免.IHA检测免疫期血清样本(1次/7d)抗体效价;二免后,FCM检测外周血淋巴细胞亚群,淋巴细胞增殖实验检测脾细胞淋巴细胞增殖情况;免疫完毕14d后用105 EID50的lasota株NDV滴鼻攻毒,统计发病率、死亡率.结果:F、HN融合基因诱导机体体液免疫和细胞免疫水平显著提高,攻毒后pVAX Ⅰ-F组保护率为30%,pVAX Ⅰ-HN组为46.7%,pVAX Ⅰ-F-HN组保护率为93.3%,表明F、HN融合基因DNA疫苗具有明显的免疫效果.

Abstract:Newcastle disease is a highly contagious disease and serious harm to the global poultry industry which caused by Newcastle disease virus. Because of their unique, DNA vaccine is safe and effective and so will be the new way to solve the shortage of traditional vaccines. To evaluate the effect of the protective antigen NDV F and HN fusion gene, PBS as control, pVAX I, pVAX I -F, pVAX I -HN and pVAX -F/ HN four kinds of plasmid were be given four 2-week-old chicks by IM and immunized twice. During the immune serum samples antibody titerwas tested once every seven days by IHA, after immunization, peripheral blood lymphocyte subsets was tested by FCM, lymphocyte proliferation of spleen ceils was tested by lymphocyte proliferation assay,14daysafter immunization, poison attack with 105 EID50 of lasota NDV and statistical the incidence and mortality. The results: the level of cellular immunity and humoral immunity were improved significantly which induction by F and HN fusion gene, the protection rates pVAX1-F group was 30%, pVAX I -HN group was 46.7%, pVAX I -F-HN group was 93.3% after NDV attack. The results proved that F and HN fusion gene DNA vaccine has obvious immune effect.
