正交试验法在牛蒡组织培养中的应用

摘    要:应用正交设计法研究BA,NAA及蔗糖3个因子对牛蒡(Arctium lappa L.)组培苗的增殖率的影响,同时考察了NAA,BA及不同浓度的Ms培养基3个因子对牛蒡组培苗生根培养的影响,各试验均按正交表L9(3^4)形成3因素、3水平的不同配比。试验结果表明:最佳的增殖培养基是MS BA2.0mg/L NAA0.05mg/L 蔗糖30g/L,最适于生根的培养基是1/2MS BA0.1mg/L NAA0.05mg/L 30g/L蔗糖。

Abstract:In order to analyze the effect of different contents of hormones and sucrose in the medium on tufted-bud induction and tufted-bud rooting of Arctium lappa L., tissue culture of the plant with the seeds as the explants was carried out, in which the orthogonal design, L_9(3~4), was used. The results indicated that MS supplemented with BA 2.0 mg/L NAA 0.05 mg/L sucrose 30 g/L was suitable for its multiplication and1/2MS supplemented with BA 0.1 mg/L NAA 0.05 mg/L sucrose 30 g/L was optimal for rooting.
