早孕小鼠子宫内膜ICAM-1的表达及意义

摘    要:探讨细胞间粘附分子-1(ICAM-1)基因及蛋白在早孕小鼠子宫内膜的表达及其意义方法:收集妊娠1-5d的早孕小鼠子宫内膜,利用RT-PCR、免疫组织化和Western-blot方法检测ICAM-1基因及蛋白的表达及分布;着床期小鼠单侧宫角注射ICAM-1单抗,观察胚胎着床变化.结果:1.ICAM-1 mRNA孕D1到D3逐渐增高,D4达最高,D5开始降低;2.子宫内膜上皮细胞,基质细胞,血管上皮细胞均有ICAM-1的表达,阳性细胞数量、表达强度和表达时间呈现一定规律性;3.ICAM-1D1、D2、D3略有升高,D4有强表达,D5略有下降;4.单侧宫角注射ICAM-1单抗后,该侧胚胎发生流产.结论:ICAM-1在早孕小鼠子宫内膜的表达变化及分布,提示ICAM-1参与了胚泡着床,ICAM-1可能起调节子宫内膜的局部免疫作用.

Abstract:Endometrium samples were collected from Day 1 to Day 5 in early pregnant mice, and the expression level of ICAM-1 (intercellular adhesion molecule-1) mRNA on it was determined by RT-PCR and the expression level of ICMA-1 were determined by immunohisto chemistry and Western blotting. Monoclone antibody of ICAM-1 was injected into the uterus on one side of the early pregnant mice to observe the changes of the embryo. The expression level of ICAM-1 mRNA on the endometrium increased steadily to Day 4 and declined gradually thereafter. Expression of ICMA-1 was detected on endometrium epithelia, matrix cells and blood vessel epithelia and the quantity of its positive cells and the intensity and duration of the expression showed a definite pattern. In Western blotting assay, ICMA-1 slightly increased from D1 to D 3 and showed a strong expression on Day 4, which was followed by a decline thereafter. The embryo aborted spontaneously on the side of the injection of monoclone antibody of ICAM-1. These results suggest that ICAM-1 may be involved in the process of blastosyst implantation and regulate the local immunity of the endometrium.
