南蛇棒魔芋的组织培养与植株再生研究

摘    要:以南蛇棒魔芋球茎、叶柄、鳞片、顶芽为外植体进行组培及再生研究。球茎、叶柄、鳞片在MS附加1.5mg/L,BA和1.5mg/L NAA的培养基中,愈伤组织诱导率迭98%;在MS附加1.5mg/LBA和0.1mg/L NAA分化培养基中.其分化率迭95%以上;纵切顶芽接入分化培养基中。可长出3个丛生芽,芽分化率迭80%以上;不定芽在生根培养基MS附加0.1mg/LBA和0.5~1.0mg/LNAA上,能100%生根形成完整的植株。

Abstract:Corms, petioles, scales and top buds of Amor phophallus dunnii Tetcher, a species of the genus Amor phophallus that is on the brink of extinction, were used as explants for tissue culture and plant regeneration. The corms, petioles and scales gave a callus induction frequency of 98% when cultured on MS medium supplemented with BA 1. 5 mg/L and NAA 1. 5 mg/L and a bud differentiation rate of more than 95% when cultured on MS medium supplemented with BA 1. 5 mg/L and NAA 0. 2 mg/L. When split top buds were inoculated on the differentiation medium, each gave rise to about 3 rosette buds, with a differentiation rate of more than 80%. On the rooting medium (MS supplemented with BA 0. 1 mg/L and NAA 0. 5-1. 0 mg/L), all the adventitious buds rooted readily and formed whole plants.
