摘    要:以5个有重要育种价值的籼型恢复系为材料,研究了影响籼稻愈伤组织诱导、生长和再生的几个重要因素。结果表明,对于籼稻品种而言,MS,NB培养基都是较为适合的培养基类型,愈伤诱导率都比较高,且愈伤质量较好,具有较强的再生能力,但是不同品种对不同培养基的反应有所不同,籼稻品种之间再生能力存在极大差异;分化培养基中适宜的激素配比为BAP3mg/L NAA0.5mg/L;适当的干燥处理可以极大地提高愈伤组织的再生能力。还从这5个品种中初步确定了3个再生频率较高的品种,作为进一步研究籼稻组织培养特性的材料,并进行下一步的遗传转化试验。

Abstract:Several factors influencing callus induction frequency, growth and plantlet regeneration were investigated, using 5 fertility-restoring lines of indica rice. MS and NB appeared to be appropriate media of callus induction, subculture and differentiation for indica rice cultivars. However, different cultivars responded differently to different media, and significant differences were present between the cultivars studied in their differentiation capacity. Suitable exogenous hormone combination for the differentiation medium was BAP 3 mg/L and NAA 0.5 mg/L. Partial desiccation of the calli before their transfer to the regeneration medium increased the frequency of plantlet regeneration significantly.
