基于SSR标记13个紫苏品种(系)的亲缘关系及遗传多样性

摘    要:为紫苏种质资源利用提供参考,利用14对SSR引物对13个紫苏品种(系)提取的DNA进行PCR扩增,并对其亲缘关系进行鉴定与遗传多样性分析。结果表明:1)从13个紫苏品种(系)中共检测出51条清晰DNA条带,其中多态性条带47条,占92.16%。2)13个紫苏品种(系)的遗传距离为0.086~0.576,平均遗传距离为0.336;遗传相似性系数为0.424~0.914,平均遗传相似性系数为0.654。3)在遗传距离(SM)为0.59时,13个紫苏品种(系)可聚为2类群。类群Ⅰ为开阳紫苏(CK)1个品种(系),类群Ⅱ包括奇苏1~6号及其原种1~6号共12个品种(系)。在SM为0.67时,类群Ⅱ又可分为2部分,第1部分包括奇苏1号和奇苏6号及其原种1号和原种6号共4个品种(系);第2部分包含奇苏2~5号及其原种2~5号共8个品种(系)。在SM为0.80时,原种2号、原种3号和原种4号的亲缘关系最近,但经系选改良后,其亲缘关系已有一定距离。

Abstract:Genomic DNA fragments extracted from 13 P.frutescens varieties are amplified by 14 pairs of SSR primers and their genetic relationship and genetic diversity are identified and analyzed to provide a reference for identification and utilization of P.frutescens germplasm resources.Results: 1) 51 distinct DNA bands are identified in 13 P.frutescens varieties.47 bands in 51 bands are of polymorphism.2) The genetic distance and average genetic distance of 13 P.frutescens varieties is 0.086~0.576 and 0.336 respectively.The genetic similarity and average genetic similarity coefficient of 13 P.frutescens varieties is 0.424~0.914 and 0.654 separately.3) 13 P.frutescens varieties can be divided into two groups when the genetic distance is 0.59.Group I includes one Kaiyang local P.frutescens variety (CK) only and Group II includes 12 P.frutescens varieties of Qisu 1~6 and Yuanzhong 1~6.Group II can be divided into two subgroups when SM is 0.67.Subgroup I includes four P.frutescens varieties of Qisu 1, Qisu 6, Yuanzhong 1 and Yuanzhong 6 and Subgroup II includes eight P.frutescens varieties of Qisu 2~5 and Yuanzhong 2~5.When SM is 0.80, the genetic relationship among Yuanzhong 2, Yuanzhong 3 and Yuanzhong 4 is more close but the genetic relationship is at a distance after systemic breeding.
