饲料中5 种糖皮质激素的超高效液相色谱检测

研究建立了一种同时检测饲料中泼尼松、甲基泼尼松龙、倍氯米松、氟氢可的松、醋酸可的松等5种糖皮质激素的液相色谱分析方法,采用乙腈作为提取溶剂,减少了杂质溶出;同时优化色谱条件,减少了背景干扰。选择Agilent Poroshell 120 SB-C18色谱柱(3.0 mm×150 mm,2.7μm),以40%乙腈水为流动相、流速0.60 mL/min、柱温35.0℃、紫外检测器(TUV)、波长254 nm进行检测,结果表明,5种糖皮质激素分离良好,6 min内即出峰完毕,线性关系良好,相关系数为0.99957～0.99996,回收率为83.80%-94.04%,检测限为0.03～0.05 mg/kg。整个试验过程只需提取、离心、吹干、定容,免除了净化步骤,达到了高效、灵敏、低消耗的效果,能满足样品分析要求。

Detection of 5 kinds of glucocorticosteroids in feed by UPLC

An analysis method was established to detect 5 kinds of glucocorticosteroids in feed by UPLC in this article.Acetonitrile was selected as extraction solvent,with less impurities dissolved out.By optimization of UPLC conditions,background interference was reduced.The Agilent Poroshell 120 SB-C18 column(3.0 mm×150 mm,2.7 μ m) was selected.40% acetonitrile-water was the mobile phase.The flow rate was 0.60 mL/min.The column temperature was 35℃.The detector was TUV,and the detection wavelength was 254 nm.The results showed that the 5 kinds of glucocorticoids were separated well,outflowing in 6 min.Good linearity was showed,with correlation coefficients of 0.99957～0.99996.The recovery rates were 83.80%～94.04%.The detection limits were 0.03～0.05 mg/kg.The entire trial process including extraction,centrifugal,blow dry and constant volume,without the step of purification,achieved the effect of high performance,flexibility,low consumption.
