空间诱变处理后菜心变异株系的RAPD分析

摘    要:为了从分子水平上研究空间诱变处理后菜心变异株系的遗传变异,对太空诱变处理后的2个菜心品种的种子返回地面进行种植,并对定向选育5~6代后的6个变异株系及原种进行RAPD检测分析。结果表明:从100条随机引物中筛选出16条能扩增出特异性条带的引物。油绿50天的3个变异株系与原种相比均有一定的遗传变异,其中以株系50-2的变异最大,3个变异株系之间有一定差异;油绿80天的3个变异株系与原种相比也有一定的遗传变异,其中以株系80-2的变异最大,3个变异株系之间也有一定差异。此外,航天诱变处理后的油绿80天菜心品种内在分子水平上的变异大于油绿50天。突变后代在DNA水平上与对照存在差异,说明突变体基因发生了点突变或染色体互换、缺失,空间诱变为菜心优良品种的选育提供了新的途径。

Abstract:To study the effects of space induction on the hereditable mutations in Brassica Parachinensis Bailey, the seeds of two Brassica Parachinersis Bailey cuhivars Youlv 50 and Youlv 80 were sent into the space by satellite to induce some mutation then sent back and planted on earth. 6 mutant lines were obtained after directive breeding for 5~6 generations. Random amplified polymorphic DNA (RAPD) method was employed to detect the genetic variation between the 6 mutant lines and un-induced control. The results ofthe amplification with 16 random primers showed that genetic variation existed between all the mutant lines of two euhivars and control in which the mutant line 50-2 and 80-2 showed the greatest variation. Intraspecific genetic variation also existed between the 3 mutant lines of Youlv 80 as well as the 3 mutant lines of Youlv 80. The intraspecific variation between 3 mutant lines of Youlv 80 was greater than that of Youlv 50. It was concluded that space mutation might provide a new route for the breeding of Brassica Parachinensis Bailey.
