非洲菊愈伤组织诱导和不定芽分化研究

摘    要:以非洲菊深圳5号为材料,研究了几种因素对非洲菊花托愈伤组织诱导和不定芽分化的影响,结果表明,花托在1/2MS 6-BA 10 mg/L NAA 0.2 mg/L IAA 0.3 mg/L培养基中诱导愈伤组织的速度最快,诱导率达100%;花托在在1/2MS基本培养基中诱导出的愈伤组织较少褐变,且易分化不定芽;将愈伤组织转接到1/2MS 6-BA 3 mg/L NAA 0.2mg/L IAA 0.1 mg/L培养基中进行继代增殖培养可有效降低褐化率,不定芽分化率达67.5%、不定芽平均数为2.3株,指出高效的愈伤组织和不定芽再生频率为非洲菊基因转化体系的建立奠定了基础。

Abstract:Effects of several factors on callus induction and adventitious bud differentiation from receptacle of Gerbera were studied.The results showed that the optimal medium for callus induction from receptacle in the primary cultivation was 1/2MS 6-BA 10 mg/L NAA 0.2 mg/L IAA 0.3 mg/L.Among 3 basic media,the 1/2MS medium was best for callus induction and adventitious bud differentiation.The suitable secondary culture medium for effective reduction of calls browning was 1/2MS 6-BA 3 mg/L NAA 0.2 mg/L IAA 0.1 mg/L,by which the differentiation rate and the average number of adventitious bud were 67.5% and 2.3 respectively.High efficient regeneration system laid the foundation for establishment of gene transformation system of Gerbera.
