应用AFLP技术分析甘蔗引进品种的遗传多样性

采用15对多态性较好的AFLP引物对36个甘蔗引进品种的遗传多样性进行分析,结果表明,每对引物的多态性条带数为45.9条、多态率为61.50%,遗传相似系数在0.6578～0.8762之间、平均为0.7459。聚类分析结果表明,36个引进品种为同一大类,而当地亲本材料粤农73-204独立成另一大类,说明粤农73-204与引进品种之间的亲缘关系较远;36个引进品种可进一步分为3类,相同系列的品种大多数能归为同一类,可见相同区域、相同机构选育的品种亲缘关系相对较近。而在系谱中亲缘关系密切的品种、大部分能归为同一类。遗传多样性指数分析结果显示,36个引进品种的遗传多样性指数为0.2470。指出加强引进品种的研究利用是促进我国甘蔗育种发展、有效提高甘蔗品种遗传多样性的重要措施。

Genetic diversity of introduced sugarcane varieties assessed by AFLP analysis

Genetic diversity in 36 introduced sugarcane varieties was studied using 15 pairs of AFLP primers.The results showed that the average polymorphic loci per pair primer were 45.9,and the percentage of polymorphism loci was 61.50%.The genetic similarity indexes of introduced varieties varied from 0.6578 to 0.8762,with an average of 0.7459.The 36 introduced varieties were clustered into one group,and YN73-204(local parent) into another group alone,which implied that the former ones were of higher genetic similarity to each other than to the latter.The former group was further divided into 3 sub-groups.Most of the varieties of same series could be classified into same sub-groups,indicating that there was a closer genetic relationship among the varieties selected by the same organization,and most of varieties with close relationship in family-tree could also be classified into same sub-groups.Genetic diversity index in the 36 introduced varieties was at low level with a value of 0.2470.The study revealed that the use of introduced varieties as parents in our sugarcane breeding program would be helpfully for broadening genetic diversity,and so facilitate the sugarcane improvement.
