细菌几丁质酶基因转化番茄的研究

摘    要:为了获得抗真菌病的番茄材料,采用农杆菌介导法将细菌几丁质酶基因转化番茄子叶,获得了抗卡那霉素的转化植株,PCR检测初步证明细菌几丁质酶基因已整合到番茄的基因组。同时对番茄耐Kan水平、转化受体的预培养以及共培后洗涤抑菌进行了研究。结果表明:卡那霉素质量浓度为50mg/L时就能完全抑制番茄子叶再生;共培后直接用抑菌剂洗涤的抑菌效果较好;番茄子叶预培养1d的转化率最高。

Abstract:For getting tomato that resists fungi diseases, a chitinase chiA gene from Serratia marcescens was transformed into tomato cotyledons by using Agrobacterium tumefaciens mediated method and transgenic plants with kanamycin resistance were obtained. The PCR examination has confirmed that the chitinase chiA gene is integrated into tomato genome. Furthermore, the anti Kan level of tomato, pre culture of cotyledons and washing cotyledons co cultured were studied. The results have shown that kanamycin at 50 mg/L can restrain tomato cotyledons regenerating completely, washing cotyledons co cultured with antibiotic controls growth of Agrobacteria tumefaciens more effectively and the highest transformation rate is got by using cotyledons pre cultured 1 day.
