苎麻叶片高效再生体系的建立

摘    要:以湘苎2号、湘苎3号、新宁青麻、四川洪县园麻的叶片为材料,构建4个品种高效、稳定的不定芽再生体系. 湘苎2号、湘苎3号、新宁青麻、四川洪县园麻最适诱导愈伤组织及分化不定芽的培养基分别为MS+0.7 mg/L TDZ+0.02 mg/L 2,4-D,MS+0.3 mg/L TDZ+0.01 mg/L 2,4-D,MS+0.3 mg/L TDZ+0.05 mg/L 2,4-D,MS+ 0.9 mg/L TDZ +0.05 mg/L 2,4-D;不定芽分化率分别为83.3%,81.7%,90.0%,86.7%;4个品种再分化时出现丛生芽,新宁青麻愈伤组织的平均再生不定芽数达2.62个,其他3个品种也都有数量不等的丛生芽产生,不定芽生根培养基为MS+ 0.05 mg/L NAA;叶块经过添加6%蔗糖培养基培养2周,再转入含3%蔗糖的培养基中培养,其中经6%蔗糖培养能显著提高湘苎2号、新宁青麻、四川洪县园麻不定芽分化率、平均再生不定芽数.

Abstract:An efficient plant regeneration systerm via adventitious shoot formation from leaf explants of ramie cultivar: Xiangzhu 2,Xiangzhu 3,Xinningqingma and Sichuanhongxianyuanma were established.The best medium for shoot regeneration of the four ramie cultivar are:Xiangzhu 2 is cultivated on MS medium supplenmented with 0.7 mg/L TDZ and 0.02 mg/L 2,4-D,Xiangzhu 3 is cultivated on MS medium supplenmented with 0.3 mg/L TDZ and 0.01 mg/L 2,4-D,Xinningqingma is cultivated on MS medium supplenmented with 0.3 mg/L TDZ...
