甘蓝型油菜种皮色泽形成与相关酶及蛋白质含量的关系

摘    要: 以3对不同来源的甘蓝型油菜黄、黑籽近等基因系为试材,研究了甘蓝型黄籽油菜种皮色泽形成与相关酶及蛋白质含量的关系。结果表明,相同遗传背景的黑籽种皮中,苯丙氨酸解氨酶(PAL) 和多酚氧化酶(PPO)均显著高于黄籽种皮,且黄籽种皮的PAL活性高峰期出现较晚,而谷氨酰胺合成酶(GS)活性在黄籽和黑籽种皮中无显著差异。在种子发育期用红光、蓝光、PAL抑制剂对羟基苯甲酸、PPO抑制剂聚乙烯吡咯烷酮、蛋白质合成促进剂尿素、质体蛋白质合成抑制剂氯霉素分别处理植株,发现PAL是黄籽种皮呈色的关键因素;PPO是黑籽种皮呈色的

Abstract:To study the relationship of the color formation and related enzymes and protein content of the spermodermsin Brassica napus, three pairs of different genetic near-isogenic lines of black-seed and yellow-seed Brassica napus wereapplied as the experimental materials in the research. The results showed that the activities of phenylalanine ammonia-lyase(PAL) and polyphenol oxidase(PPO) in black-seed lines were much higher than those in yellow ones under the same geneticconditions. Additionally, the peak of the PAL in yellow-seed spermoderms appeared comparatively late. However, theactivities of glutamine synthetase (GS) in yellow-seed and black-seed spermoderms presented no apparent difference.During the seeds developing, the plants of yellow-seed and black-seed Brassica napus were treated with red light, bluelight, p-thydroxybenzoic acid PAL-inhibiter, polyvinylpyrrolidone PPO-inhibiter, urea the promoter of protein synthesisand chloramphenicol the inhibiter of plastid protein synthesis, respectively. It was found that PAL and PPO might be the keyfactors to make the yellow-seed and black-seed spermoderms color respectively. Nitrogen assimilation might have no directrelation with the difference of the protein content of yellow-seed and black-seed spermoderms. PAL was the essential factorto make the protein content of yellow-seed and black-seed spermoderms different.
