基于PCR技术的玉米丝轴黑粉菌侵染率

摘    要:摘要:以对丝黑穗病高感的黄早四和高抗的Mo17自交系为材料,用丝轴黑粉菌进行接种,采用丝黑穗病病原菌基因组特异引物的PCR技术,分别对幼苗期的根,茎,叶和成熟期的根,茎,叶,雄穗DNA进行PCR扩增,研究病菌对玉米的侵染率及扩展进程。结果表明;特异引物的PCR技术不仅能够对玉米幼苗是否受到病菌的侵染进行早期准确鉴定,而且对丝轴黑粉菌在体内的扩展进程的跟踪研究证明,玉米对丝黑穗病的抗性差异主要表现为抗侵染和抗扩展,抽穗期的雄穗对病樱的形成不能表现出明显的抗性效应。

Abstract:Objective] Head smut of maize substantially reduces grain yield. Here we demonstrate a recent survey of the rate of infection of head smut by PCR. Method] Using gene-specific primer derived from the head smut genomics and maize temporal and tissue-specific DNA, the invading process of the pathogen is displayed clearly. Result ] The study shows that the differential resistance of maize against head smut depends mainly on the ability of resistance against infection and spreading. Conclusion ] The above effect disappears in the stamen when the tumor forms during tasseling stage.
