小麦愈伤组织状态调控与原生质体培养

摘    要: 愈伤组织及其细胞的状态可通过调整培养基成分来调控。一般情况下,生长素、还原态氮、KCl促进愈伤组织生长,细胞分裂素、硝态氮抑制愈伤组织生长。绝大多数易培养基因型诱导的愈伤组织往往表现为质地紧硬或外软内硬,不适于进行悬浮培养和原生质体培养。利用不同水平的2,4-D、NH#++#-4、谷氨酰胺、水解酪蛋白、KCl、KT、6-BA、NO#+-#-3等进行处理,可使不适的愈伤组织变成适宜的类型,也可使那些生长过旺而不分化的愈伤组织分化出植株。用这些方法,从6个小麦基因型的原生质体中培养出了再生植株。

Abstract:The status of wheat calli in tissue culture can be regulated by adjusting the compositions of cultural media. Generally, auxin, reducing N and KCl stimulate the growth of calli whereas cytokinin and NO - 3 show inhibitory effects on calli. Most of the calli easily obtained have their texture too compact or compact inside but soft outside that are not suitable for suspension culture and protoplast culture. However, they can be transformed into suitable types for suspension and protoplast culture by treating them with 2,4 D, NH 4,glutamine,casein hydrolysate,KCl,kinetin, 6 BA and NO - 3 at various concentrations. Procedures were set up for regulating calli to be vigorous for differentiation. By those techniques, plantlets were regenerated from protoplast culture of 6 wheat genotypes.
