快生型大豆根瘤菌G+Cmol%测定及与其它根瘤菌之间的DNA同源性

摘    要:用热变性温度法测定了快生型大豆根瘤菌DNA中G+Cmol%,结果表明其C+Cmol%为58.4-65.6,菌株间差为7%,提示了不同种的存在。以液相复性速率法测定了快生型大豆根瘤菌与其它根瘤菌种之间的DNA同源水平,测定证明它们之间的同源水平很低。快生型大豆根瘤菌与慢生型大豆根瘤菌三个DNA同源组间的同源率分别力15,10和25%,表明其亲缘关系很远。同源水平的测定为进一步研究奠定了基础。

Abstract:The G+C mol% of the DNA and deoxyribonucleic acid homology were determined between fast-growing soybean nodulating bacteria from Liaoning province and the other rhizobia by thermal denaturation temperature (Tm) and liquid phase chromatogram respectively. The results show that the G+C mol% of the DNA for these strains is 58.6-65.6%,but most of them are between 61-65%, difference among the strains is 7%. The mean level of the DNA homology of 45 hybridizations is very low with the representation strains of 3 homology groups for B. japonicum and reference strains of B. sp (cowpea) strain 8B6, R. phaseoli (127K17), R. leguminosarum (ATCC_(10004), R.loti (B), R. trifolii (540-80), R.meliloti (ATCC_(9930). The results Provided a base for further classifying fast-growing soybean rhizobia.
