肉鸡肥度性状的RAPD标记研究

 供试鸡为30只AA肉仔鸡，其中公、母仔鸡各15只。测定49日龄活重、屠体重、腹脂重、腹脂率、肝脏鲜重、干肝脂肪含量、胸肌干样脂肪含量和血浆VLDL浓度。采血分离出DNA后，对公、母鸡群分别利用已筛选出的17个随机引物分别对所有个体的基因组进行PCR扩增。对多态性片段，将“有带”、“无带”个体组成的群体看作是独立样本，比较两样本间数量性状的差异。结果在公、母鸡群分别发现了29、35个显著关联的标记与性状组合。经对与同一标记同时关联的性状间的相关分析及两样本间个体表型值分布的分析，发现上述大部分标记与性状关联是不可信的。在公鸡群没有发现显著的标记与性状关联，在母鸡群检测到两个显著的标记与性状关联，它们是OP1308-腹脂重和OP1308-腹脂率。

Study on the RAPD Markers of Fatness Traits in Broiler Chickens

The 15 male and 15 female broilers available for test were from a commercial line of broiler chicken(AA). At 49 d of age, live weight, dressed weight, abdominal fat weight, percentage of abdominal fat, fresh weight of liver, fat content of dry liver, fat content of dry breast muscle and plasma VLDL concentration were measured. Blood was sampled from individuals and the DNA isolated from the blood was used to screen for RAPD fingerprint bands that associated with the phenotypic traits. To identify linkage of bands with QTL, 17 arbitrary primers selected previously were used in the male and female populations respectively, and difference between the quantitative characteristics of two groups, according to the present or absent of bands, was compared. Twenty nine fingerprint bands were identified to be linked with QTL of specific traits in male population, and 35 in female population. It was found that most of these associations between QTL and genetic maker were unreliable through the analysis of correlation between traits that were linked with the same band and the comparison of phenotypic distribution of traits between two groups. There was no reliable markertrait linkages in male population, and two ones, OP1308abdominal fat weight and OP1308the percentage of abdominal fat, were found in female population.
