| 摘 要： | PCR扩增猪圆环病毒2型(PCV2)的衣壳蛋白(Capsid)基因,鉴定后克隆到杆状病毒转移载体pFastBacTM1中,获得重组转移载体pFastBac-Cap,转化进含穿梭载体Bacmid的感受态细胞DH10Bac中,经转座作用,蓝白菌落筛选得到含cap基因的重组穿梭载体Bacmid-Cap,以脂质体介导的方法将重组穿梭载体转染Sf9昆虫细胞,获得重组病毒reBacCap,PCR能够扩增出相应大小的片段,间接免疫荧光分析表明,PCV2阳性血清能使reBac-Cap感染的Sf9昆虫细胞出现特异性绿色荧光,表明reBac-Cap在Sf9细胞中成功表达PCV2-Cap蛋白.动物试验表明该重组病毒对小鼠有一定的保护力.

| Abstract: | The capsid protein gene of porcine circovirus type 2(PCV2) was amplified by PCR and cloned into baculovirus transfer vector pFastBacTM1.Confirmed by sequencing,the vector pFastBac-Cap was transformed into E.coli DH10Bac competent cells containing baculovirus shutter vector(Bacmid) and helper vector.Within E.coli DH10Bac,the cap gene was transposed into the bacmid.The positive white colonies of E.coli containing recombinant bacmid(Bacmid-Cap) were selected.The Bacmid-Cap was transfected into Sf9 cells to yie... |
