| 摘 要： | 采用RT-PCR扩增出鸡传染性支气管炎病毒(IBV)M 41株的全长S1基因,并将其片段连接到pGEM-T载体,经酶切、PCR及序列测定表明,获得的S1基因ORF全长为1 659 bp,与G enB ank上公布的IBV M 41 S1基因序列一致。将S1基因与真核表达质粒pVAX 1连接,构建重组真核表达质粒pVAX 1-S1。将重组质粒转染COS-7细胞,采用间接免疫荧光检测转染细胞S1蛋白的表达。同时以重组质粒免疫小鼠,可诱导产生特异性针对IBV S1蛋白的抗体(P<0.01)。这为鸡传染性支气管炎新型基因工程疫苗的研制奠定了基础。

| Abstract: | The complete S1 gene from strain M41 of infectious bronchitis virus(IBV) was amplified by RT-PCR and cloned into the pGEM-T vector.After confirmed by the restriction endonuclease analysis and PCR assay,the positive clone of S1 gene was sequenced and then transferred into eukaryotic expressing vector pVAX1.The recombinant plasmid pVAX1-S1 was transfected into COS-7 cells.The expressed S1 protein were detected with indirect immunofluorescence assay.The recombinant plasmid pVAX1-S1 could significantly elicit specific antibody response in immunized mice compared with pVAX1(P<0.01).These results lay foundation for developing the IBV DNA vaccine. |
