正交设计龙胆提取工艺的研究

摘    要:目的建立龙胆最佳提取工艺,为龙胆临床及药理研究奠定基础。方法试验采用超声提取法,以溶剂浓度(A)、超声时间(B)和料液比(C)为三因素,设计正交试验,优化龙胆提取工艺;同时,应用HPLC测定各组合龙胆苦苷含量。结果影响因素最大的是溶剂浓度,其次是超声时间,影响最小的是料液比。结论各因素较优的水平组合为A2B3C1,即用乙醇浓度为50%、超声时间为70min、料液比为1:4的条件下,提取龙胆得膏率最高。

Abstract:Objectivev For further researching on the clinic and pharmacology action of G.scabra Bge,it Was founded for the best extracting craftwork of G.scabra Bunge.Methods the study applied on super-sonic extracting method.we designed orthogonal experiment by three facts for example solvent concentration(A),super-sonic time(B),liquid-material ratio(C).Gentiopicrin content was detected with HPLC.Result the solvent concentration(A) is the most effective factor.the second is super-sonic time(B).the last is liquid-material ratio(C).Conclution the best extracting craftwork is A2B3C1 : ethanol concentration50%,super-sonic time70min,material-liquid ratio1:4.
