摘    要:以无菌萌发罗汉果子叶为外植体,进行罗汉果高效离体再生体系的研究。探讨了不同MS、琼脂含量、不同激素浓度配比、种子萌发时间和活性炭在罗汉果种子萌发、子叶诱导不定芽及试管苗生根方面的影响。结果表明:1/4 MS、0.5%-0.6%的琼脂含量比较适合罗汉果种子萌发,萌发率达89.2%;降低无机盐和蔗糖浓度有利于罗汉果试管苗的生根;最适生根培养基为1/2 MS+IBA0.5mg/L+1.5%蔗糖;罗汉果子叶直接诱导不定芽最适合培养基为1/2MS+6-BA1.0mg/L+IBA0.1mg/L,分化率达94.8%,平均每块子叶出芽10个;培养基中添加低浓度(0.2%)活性炭有利于生根,但不利于子叶不定芽的诱导;子叶不定芽的分化率还受种子萌发时间的影响,萌发时间以2—4d为宜;试管苗经过炼苗2~4d能极大提高移栽成活率,达91.7%。

Abstract:The study was carried on tissue culture of cotyledon for the establishment of effective regeneration system of Siraitia grosvenirii. The effects of different MS, Agar concentration, hormone combinations, seed germination time and active carbon (AC)on induced adventitious buds via cotyledon and the propagation in vitro were investigated. The results showed that 1/4 MS with 0.5-0.6 % Agar was optimum for seed germination, and the germination rate was 89.2 %. It is in favor of rooting to reduce the concentration of inorganic salts and sucrose, and best rooting medium was 1/2 MS + IBA0.5 mg/L+ 1.5 % sucrose; the medium of 1/2 MS+6- BA1.0 mg/L + IBA0.1 mg/L was the most suitable for inducing adventitious buds of cotyledon. Differentiation rate was 95.8 % with an average of 10 adventitious buds per cotyledon;to add low concentration (0.2 % )of AC in medium was conducive to rooting, but not conducive to adventitious buds of cotyledon. Seed germination time affected the differentiation of adventitious buds, germination time for 2-4 days is more suitable for adventitious buds differentiation; after hardening, the survival rates of tube plantlets were improved to 91.7 %.
