金花茶体胚和叶片愈伤组织培养

摘    要:分别以金花茶子叶胚切块和成年植株叶片为材料,进行愈伤组织培养的研究。结果表明:金花茶子叶胚去外表皮后切块,接种于MS+1.5mg·mL-12,4-D+0.5mg·mL-1KT中,能诱导出愈伤组织,并能较好地继代,蔗糖浓度为6%时,愈伤组织生长良好;金花茶成年叶片在流水中冲洗30min,75%酒精浸泡30s后,转入0.2%HgCl2消毒剂中浸泡8min,接种于MS+1.5mg·mL-16-BA+0.5mg·mL-1IAA+4mg·mL-1NAA中,愈伤诱导率为98.335%,遮光有助于叶片愈伤组织的生长。

Abstract:In this experiment,cotyledonary somatic embryos and the leaves from the adult plant were used as the materials for callus culture in Camellia nitidissima Chi..The results indicated that the pieces cut from cotyledonary somatic embryos were cultured on the MS medium with 1.5 mg·L-1 2,4-D and 0.5 mg·L-1 KT,callus could be induced and subcultured well,and at 6% sucrose concentration the callus grew well.The adult plant leaves were washed in water for 30 minutes,before putting in 70% ethanol for 30 s.Then the adult plant leaves were surface-disinfected for 8 minutes in HgCl2(0.2%).This kind of treatment could achieve good sterilation effect.The pieces cut from the adult plant leaves were cultured on the MS medium with 1.5 mg·L-1 6-BA,0.5 mg·L-1 IAA and 0.5 mg·L-1 NAA,callus could be induced.The highest frequency of callus induction was 98.335%,shading was helpful to the growth of callus.
