金樱子槲皮素提取工艺的优化

摘    要:为了优化金樱子(Rosa laevigata Mickx.)槲皮素的提取工艺,以槲皮素提取率为指标,考察提取温度、超声时间、乙醇体积分数、料液比、提取次数5个单因素对金樱子槲皮素提取率的影响,又采用正交试验优化提取工艺.结果显示,优化的金樱子槲皮素的工艺条件为乙醇体积分数60%,提取温度70℃,料液比1∶20 (m/V,g∶mL),超声时间60 min,提取3次;各因素对金樱子槲皮素提取率的影响为乙醇体积分数>提取温度>料液比>超声时间.在优选的工艺条件下金樱子槲皮素的提取率为2.735%,高于正交试验中的最高提取率,表明通过正交试验达到了优化目的,优选的最佳提取工艺条件稳定可靠.

Abstract:In order to optimize the extraction technology of quercetin from Rosa laevigata Mickx.,the yield of quercetin was took as the index,and single-factor experiments were adopted to study the effects of extraction temperature,ultrasonic time,volume fraction of ethanol,solid to liquid ratio and extraction times on the yield of quercetin,and orthogonal design was used to optimize the extraction technology.Results showed that the optimal processing conditions were,volume fraction of ethanol,60%;extraction temperature,70 ℃;solid to liquid ratio,1∶20(m/V,g∶mL);ultrasonic time,60 min;extracting for 3 times.Factors affecting the yield of quercetin ranked as volume fraction of ethanol>extraction temperature>solid to liquid ratio>ultrasonic time.Under the optimal conditions,the yield of quercetin reached up to 2.735%,which was higher than any of the treatment of orthogonal test,indicating that optimization of the extraction process had been achieved;and the obtained process was reliable and stable.
