辅以多粘类芽孢杆菌堆肥提取液工艺及其对土壤微生物群落结构的影响

摘    要:以猪粪堆肥为原料,建立多粘类芽孢杆菌(Paenibacillus polymyxa)与猪粪堆肥提取液的配伍技术工艺,并研究其对烟草黑胫病(Phytophthora parasitica)土壤微生物群落结构的影响。结果表明,最佳技术工艺是多粘类芽孢杆菌添加到猪粪堆肥中发酵72 h,再按照水∶堆肥8∶1(质量比)浸提48 h。不同工艺提取的猪粪堆肥提取液均能有效抑制烟草黑胫病病原菌的生长。与对照相比,施用辅以多粘类芽孢杆菌的堆肥提取液处理土壤疫霉ITS基因拷贝数减少了18.15%~53.33%,土壤中芽孢杆菌数量增加了45.63%~255.00%,同时增加了土壤细菌和放线菌数量,但是减少了真菌数量。辅以多粘类芽孢杆菌的猪粪堆肥提取液处理增加了Bacillus niabensis和B.aryabhttai的丰富度和细菌遗传多样性,但降低了真菌的丰富度和遗传多样性。

Abstract:The pig manure carried out to established the compatible technology of Paenibacillus polymyxa and pig manure compost extracts,and the effect of pig manure compost extracts combined with P. polymyxa on soil microbial community structure was studied in Phytophthora parasitica. The results showed that the best compatible technology was that the pig ma-nure added P. polymyxa ferment for 72 h,and 1 kg of compound pig manure with P. polymyxa mixed with 8 kg water (weight ratio),and then extracted for 48 h. All compost extracts which extracted with different technology could inhibit the growth of P. parasitica. When compared to water application treatment, the internal transcribed spacer (ITS) gene copy number de-creased by 18.15%~53.33% in pig manure compost extracts combined with P. polymyxa treatment,and soil bacillus number increased by 45.63%~255.00%. The bacteria and actinomyces number of pig manure compost extracts combined with P. polymyxa treatment were increased, but the number of fungi was decreased. The richness of Bacillus niabensis and B. aryab-httai, and the diversity of bacterial community structure were all improved, however, the richness of fungus and the diversity of fungus community structure were all decreased,when the soil applied with pig manure compost extracts combined with P. polymyxa.
