| 摘 要： | 根据已报道的DsMV中的衣壳蛋白(cp)基因序列,首先合成1对扩增DsMV的cp基因中间部分核心序列的寡核苷酸引物;然后将提取的魔芋植株的总RNA反转成cDNA,再以cDNA为模板,用上述引物进行常规PCR反应,结果扩增得到了与预期大小一致的0.35kb片段,健康魔芋植株接种DsMV后也扩增得到同样大小的片段,而健康植株则未扩增到任何片段。从而建立了魔芋中DsMV经济简便的RT-PCR检测体系,为魔芋DsMV的防治和脱毒苗的检测提供了有效手段。 |

| Abstract: | A pair of primers was synthesized based on the previously reported primers designed for detecting DsMV based on the core nucleotide sequence of the cp gene of dasheen mosaic virus(DsMV)genome.The fragment of cp gene of DsMV was amplified by RT-PCR according to two differential procedures respectively.The target fragment about 0.35kb was amplifled from the infected samples and inoculated samples but not from health sample.The economic and simple method of detecting DsMV affecting konjak was built and could be used in preventing the spread of DsMV and detecting the seedling wether it was detoxicated or not. |
