生物酶法提取沙棘果渣总黄酮工艺的优化

摘    要:用生物酶法提取沙棘(Hippophae rhamnoides)果渣中的总黄酮,通过单因素试验和正交试验优化提取工艺。结果表明,酶解体系各因素对总黄酮提取率影响的大小顺序为酶用量、酶解温度、酶解时间、pH,最佳提取条件为生物酶用量占沙棘果渣质量的4%、pH 5、酶解温度60℃、酶解时间120 min,料水质量比1∶30,此条件下总黄酮提取率可达到0.796%。与有机溶剂回流法和超声波法相比,生物酶法所得沙棘果渣的总黄酮提取率分别提高了51.91%和48.51%。

Abstract:Bio-enzyme method was applied for total flavones extraction from marc of sea buckthorn(Hippophae rhamnoides);and the extraction craft was optimized by single factor experiments and orthogonal design.The results showed that the optimum extraction conditions for the enzyme reaction system were as follows,enzyme dose,4% of the mass of marc of sea buckthorn;pH,5;Temperature,60 ℃;Time,120 min;Mass ratio of material to water,1∶30.Under the optimum conditions,the yield of total flavones was up to 0.796%.Compared with heat reflux extraction and ultra-sonic extraction,the yield of bio-enzyme extraction was increased by 51.91% and 48.51%,respectively.
