耐高温纤维素酶产生菌的筛选鉴定及酶谱分析

摘    要:采用稀释平板-刚果红染色法从腐木中分离到1株产耐高温纤维素酶的菌株CY15-10.通过形态观察、特征培养及16 S rDNA序列分析,鉴定该菌株为多粘类芽孢杆菌(Paenibacillus polymyxa).该菌株经摇瓶发酵后测定粗酶液CMC酶活为1.27 U/mL,最适反应温度为55℃,热稳定性较好.经聚丙烯酰胺凝胶电泳(SDS-PAGE)及酶谱分析表明该菌株向胞外分泌1种纤维素酶组分,酶分子质量约为35 ku.

Abstract:A hyperthermophilic cellulase-producing strain CY15-10 was isolated from decayed wood and compost through the congo red medium plate method.Based on morphology,biochemical and physiological characterization,16 S rRNA sequence analysis and construction of phylogenetic tree,CY15-10 strain was identified as Paenibacillus polymyxa.The CMCase was 1.27 U/mL.The enzyme had the highest activity when temperature was 55 ℃ and performed good thermal stability.The SDS-PAGE and zymographic analysis detected that a peptide with molecular mass of 35 ku was excretived.
