桦褐孔菌多糖的提取及含量测定方法的研究

摘    要:采用水提醇沉法提取桦褐孔菌粗多糖,并利用苯酚-硫酸比色法,以葡萄糖为标准品,测定桦褐孔菌菌核及其粗多糖中多糖的含量.结果表明,桦褐孔菌粗多糖得率为17%,桦褐孔菌菌核中多糖含量为2.068%,桦褐孔菌粗多糖中的多糖含量为22.43%.该方法桦褐孔菌粗多糖得率高,加样回收率在99%以上,且5 h内显色稳定,重现性好,适合用于中药桦褐孔菌多糖的提取及含量测定.

Abstract:The contents of polysaccharides in sclerotium(Inonotus obliquus)and its crude polysaccharides were detected by the method of phenol-vitriolic colorimetry taken glucose as standard after water extraction and alcohol precipitation.The results indicated that the crude polysaceharides in Inonotus obliquus,polysaecharides in sclerotium and polysaceharides in crude polysaceharides were 17%,2.068% and 22.43% respectively.In a conclusion,water extraction and alcohol precipitation were effective methods to obtain high yield of polysaceharide from Inonotus obliquus,and phenol-vitriolic colorimetry was suitable for polysaccharide determination for its high recovery (>99%),stabihty (in 5 hours),repeatability and accuracy.
