中国沙棘染色体的染色和制片技术

摘    要:以中国沙棘的茎尖为材料,进行不同的取样时间、预处理、固定、解离时间处理后,压片观察染色体。结果表明:早上09:00~10:00取生长旺盛的茎尖组织,用8-羟基喹啉预处理材料1.5~2.5h,卡诺固定液固定2~3h置于60℃恒温水浴酸化15~20min,用改良的卡宝品红染色15min,制出的玻片染色体清楚,效果极佳。

Abstract:Chromosome observation on different material sampling time, pretreatment time, fixation time, acidification time were studied. The results showed that the clear chromosome was observed with the materials being treated by means of selecting eselect tissue of threvs stem tip at 09:00 - 10:00 in the morning, being immersed between one and a half hours and two and a half hours with 8 - hydroxyquinoline, fexing between two hours and three hours with kanuo stationary liquid, acidating between fifteen minutes and twenty minutes, dying in fifteen minutes with improved carbor fuchsin.
