质粒DNA提取与酶切方法的比较研究

摘    要:比较了质粒DNA小量和大量制备方法,分析了质粒纯度、酶的种类、单酶切和双酶切方法对酶切效果的影响。结果显示,质粒DNA小量制备法效果好,提取的质粒量大,质量高;GalUR/TEM-T质粒双酶切时需先经过纯化才能酶切完全;BamHI单酶切FRO2/PCB302质粒效果比较好,而XbaI单酶切FRO2/PCB302质粒效率比较低,需要纯化和降低酶切反应中质粒的浓度。

Abstract:To distill plasmid DNA effectively,two methods were compared(small and large quantity).The enzyme-cut efficiency was researched by plasmid purification,single or double enzyme-cut method,different kind of enzyme.The result showed that small quantity method was better with high quality and quantity.After purified the GalUR/TEM-T vectors,their enzyme-cut reaction was completely.The enzyme-cut efficiency of FRO2/PCB302 by BamHI was better than XbaI.If XbaI enzyme cut FRO2/PCB302,the plasmid should be purified at first step and lower the content.
