四种当归多糖提取方法的比较研究

摘    要:对热水浸提法、超声波法、纤维素酶与果胶酶法这4种提取当归多糖的方法进行比较.以当归多糖得率为指标,用苯酚-硫酸法对当归多糖进行测定,优化4种方法的提取工艺并比较.结果显示,在最佳工艺条件下,热水浸提法的多糖得率为21.78%,超声波法为22.15%,纤维素酶法为24.55%,果胶酶法为24.81%.酶法是当归多糖提取较佳的选择.纤维素酶法的最佳提取条件是加酶量1.0%,提取温度60℃,提取时间60 min,pH值6.0;果胶酶法的最佳提取条件是加酶量1.0%,提取温度50℃,提取时间90 min,pH值5.5.

Comparison on Four Extraction Methods of Polysaccharide from Angelica Sinensis

Abstract:To compare four extraction methods of polysaccharide from Angelica s inensis, which were hot water method,ultrasonic wave method,cellulase method and pectinase method.Using the yield of polysaccharide as index, measuring the polysaccharide content by the phenol-sulfuric acid method,these four methods were optimized and compared. Under the optimal conditions,the yield of polysaccharide from Angelica sinensis was 21.78% in hot water process,22.15% in ultrasonic process,24.55% in cellulase process,and 24.81% in pectinase process,respectively.The best extraction process of Angelica sinensis polysaccharide was enzymatic method.The optimal extraction conditions of cellulase method were as follows:enzyme dosage 1.0%,at pH 6 and 60℃ for 60 min, and those of pectinase method were : enzyme dosage 1.0%, at pH 5.5 and 50℃ for 90 min.
