非洲紫罗兰组培技术研究

摘    要:以非洲紫罗兰幼嫩叶片为外植体,用MS作基本培养基,添加不同浓度BA、NAA、GA等探讨对其愈伤组织诱导、不定芽分化、继代增殖及不定根形成的影响,并筛选了适宜的激素水平范围。试验表明:采用MS+0.1~2.0m gL/6-BA+0.01~0.5mgL/NAA培养基,愈伤诱导率在20%以上;采用M S+0.1~1.0mgL/6-BA+0.01~0.5mgL/NAA+0.1~1.0mgL/GA培养基对芽的生长及增殖效果好,并提出GA具有提高不定芽长势和成苗率的作用;采用1/2M S+0.1~0.5mgL/NAA或0.1mgL/IBA或0.1~0.5mgL/IAA,20d内生根率可达97%以上。在腐叶土∶珍珠岩=5∶3的基质中驯苗,成活率可达90%以上。

Abstract:With saintpaulia ionatha wendl leaflet as explants and MS as basic culture medium,multiplication cultivation and culturing seedings of saintpaulia ionatha wendl were tested in 6-BA?NAA?GA at different density.While MS+0.1~2.0 mg/L BA+0.01~0.5 mg/L NAA were applied as culture medium,the rate is greater than 20%.In MS+0.1~1.0 mg/L 6-BA+0.01~0.5 mg/L NAA+0.1~1.0 mg/L GA medium,it had good multiplication effect and growth of sprouts.GA promoted indefinite bud growth and seeding rate.With medium of 1/2MS+0.1~0.5 mg/L NAA or 0.1 mg/L IBA or 0.1~0.5 mg/L IAA makes the rooting rate of 97% in 20 days;the survival rate would exceed 90% when the sprouts were transplanted into the 5∶3 humus soil and perlite substrate.
