摘    要:以甘蓝型油菜单倍体试管苗或花粉胚状体作为试验材料进行遗传转化。结果表明,花粉胚状体在液体培养基加入2,4-D1.0mg·L-1预培养6d,得到的绿色愈伤率最高;甘蓝型油菜花粉植株PCK121茎段在预培养基(6-BA0.5mg·L-1+2,4-D2.0mg·L-1)上预培养后得到的绿色抗性愈伤率最高;油菜单倍体试管苗叶柄和茎段在预培养时间0~10d后转化均可产生抗性愈伤;在培养基中添加2,4-D时,很容易产生绿色抗性愈伤,这是因为此时培养物对卡那霉素的耐受性增加。

Abstract:Brassica napus haploid test-tube plantlets or pollen embryoids were used as materials for transformation. The results showed that pollen embryoids precultured in liquid medium by adding 2,4-D 1.0 mg·L-1 for six days had the highest green callus percentage. The stem of the pollen-derived plantlets PCK121 got a maximum green callus percentage in meduim with 6-BA 0.5mg·L-1+2,4-D 2.0 mg·L-1. The leave petiole and shoot-stem transformed after 0~10 days preculture could produce resistent callus. Resistent callus could be easily obtained when the acceptor were cultured in medium with 2,4-D, which suggested that the cultures had increasing resistence to kanamycine.
