草坪植物RNA提取方法的比较研究

摘    要:用CTAB法、SDS法、快速SDS法、异硫氰酸胍法、Tris-硼酸法和Trizol一步法等6种方法.分别从草坪植物草地早熟禾(Poapratensis L.)、高羊茅(Festuca arundinacea Schreb.)以及马蹄金(Dichondra repens Forst.)叶片中提取总RNA.并比较各种方法提取RNA的质量。结果表明,由异硫氰酸胍法、SDS法和Tris-硼酸法3种方法获得的RNA纯度较低,存在降解和弥散现象,或混杂的DNA、蛋白质较多,效果均不理想;而用CTAB法、快速SDS法和Trizol试剂盒提取的RNA很少有降解.所得的28SrRNA和18SrRNA条带十分清晰.且D260/D280在1.871至2.021之间。快速SDS法是一种从草坪植物中有效提取高质量RNA的方法.在完整性、纯度、产率上均优于其他几种方法。

Abstract:In order to isolate high quality RNA from Poa pratensis L.,Festuca arundinacea Schreb.and Dichondra repens Forst.,several methods such as CTAB method,rapid SDS method,SDS method,guanidine isothiocyanate method,Tris-borated method and Trizol regent were used.The quality of total RNA is analyzed through gel electrophoresis and UV spectrometer.The results indicated that all the RNA isolated by guanidine isothiocyanate method,SDS method and Tris-borated method were degraded and dispersed in some degrees.RNA isolated by CTAB method,rapid SDS method and Trizol regent showed clear bands of 28S rRNA and 18S rRNA,and the value of D260/D280 is from 1.871 to 2.021.Due to fewer steps,less time,lower price and suitable for large quantity samples,rapid SDS method was an desirable method to extract total RNA from different turfgrass plant.
