摘    要:该研究以毛白杨及其杂种毛新杨和银白杨与腺杨的杂种银腺杨为材料 ,利用控制授粉杂交和种子组织培养技术相结合制备了 3个规模较大的遗传作图群体 .在此基础上 ,采用扩增性片段长度多态性 (AmplifiedFragmentLengthPolymorphisms,AFLPs)标记技术对这些作图群体的亲本进行了多态性检测 ,结果表明 :8对EcoRⅠ MseⅠ引物组合在毛新杨无性系TB0 1×毛白杨无性系LM5 0作图群体亲本间的多态性水平最高 ,平均可检测到 2 0个多态性位点 .结合对子代在田间表型性状分离情况 ,选取了该分离群体作为构建毛白杨遗传连锁图的作图群体 .并利用AFLP技术对随机抽取的 12 0株子代个体进行了基因型检测 ,10对AFLP引物组合在作图亲本间共检测到 197个多态性位点 ,其中有 15 8个位点在P <0 0 1水平上符合 1∶1孟德尔期望分离比 ,占多态性位点总数的 80 2 % ,选取该群体作为构建毛白杨遗传连锁图的作图群体是合适的

Abstract:The three large mapping populations for Populus tomentosa have been established by combining the controlled hybridization and seed culture \%in vitro,\% using the material of Populus tomentosa, P. tomentosa×P. bolleana or P. alba×P. glandulossa. Based on it, the authors have detected the levels of polymorphisms between parents of three mapping population employing the 8 AFLP primer combinations. On average, each primer pairs could produce most informative polymorphism loci of 20 between the hybrid clone “TB01” (P. tomentosa×P. bolleana) and clone “LM50” (P. tomentosa) among the parents of three mapping populations. Therefore, the mapping population of “TB01”דLM50”, considering the segregation of phenotypic traits, was selected for the construction of the first genetic linkage map in P. tomentosa. The authors further investigated segregation of genotype in 120 progeny of selecting mapping population using AFLP marker. A total of 197 polymorphic fragments were detected with 10 Eco RⅠ/ Mse Ⅰ primer combinations, among which 158 (80 2%) segregated in a expected Mendelian 1∶1 ratio ( P < 0 01), corresponding to DNA polymorphisms heterozygous in one parent and null in the other. The results show that the mapping population of “TB01”דLM50” was optimal for construction of genetic linkage map in P. tomentosa.
