摘    要:该文使用AFLP标记法技术 ,以半同胞家系的‘早凝馨’、‘凝馨’ ,全同胞家系选育出的品种‘美人’梅、‘小美人’、‘俏美人’、‘黑美人’和朱砂型相近梅花品种‘徽州骨红’、‘舞朱砂’、‘小骨里红’共计 9个品种为样本 ,选用 7对引物组合E ACT M CAT、E GA M CTC、E ACT M CCA、E ACT M CAC、E ACT M CAG、E ACC M CAG和E ACT M CTC扩增得到 2 97个多态性位点的条带数据 ,分别选用简单匹配系数和Nei(72 )遗传距离系数 ,进行不加权成对算术平均法聚类分析 ,分别建立了聚类树 .分析结果表明 :AFLPDNA指纹灵敏地鉴别相近品种 ,并与形态学鉴别分类结论相吻合 ,表明DNA指纹鉴定配合形态学鉴定方法具有较高的准确性和有效性 ;聚类分析显示了供试品种间的亲缘关系 ,朱砂型组的 3个供试品种具有比前两个家系更近的亲缘关系 .

Abstract:The amplified fragment length polymorphisms (AFLP) technique was used to assess the genetic relationships in 9 cultivars representing the three styles of Prunus mume , four cultivars of full_sib pedigree from ‘Meiren' style, two half_sib cultivars of ‘Gongfen’ style and three cultivars of ‘Zhusha’ style. Seven AFLP primerpairs, E ACT/M CAT, E GA/M CTC, E ACT/M CCA, E ACT/M CAC, E ACT/M CAG, E ACC/M CAG and E ACT/ M CTC, produced 297 polymorphic bands among the cultivars. Two dendrograms were generated by the unweighted pair_group method of arithmetic averages (UPGMA) analyses using simple matching coefficients (SM) and Nei(72)distance coefficients, producing three clusters that closely corresponded with the genetic relationships based on morphological traits. All the cultivars included are identified from each other. The two pedigree, and one style studied falls in to three distinct clusters. Three cultivars of ‘Zhusha’ style in the assay have closer genetic relationships than other sibs.
