摘    要:用 8 0个 10bp随机引物对我国新疆核桃、华北山地核桃、秦巴山地核桃和西藏高地核桃 4个不同地理生态型的 2 0个样本进行RAPD扩增反应 .选用了清晰度高、重复性好的 2 4个引物用于进一步的分析 ,共扩增出 2 2 7个RAPD片段 ,具多态性的片段有 16 8个 ,占总扩增片段的 74% ,平均每个引物扩增片段为 9 46个 ,扩增片段长度为2 2 0~ 40 72bp .其中 ,西藏核桃类群的特有RAPD片段占有很大比例 .依据扩增结果 ,计算遗传距离 ,采用UPGMA方法构建聚类树状图 .结果表明 :新疆核桃为我国栽培核桃的一个地理生态型 ;华北山地核桃和秦巴山地核桃应属一个地理生态型下的两个不同亚群 ;西藏高地核桃不仅在聚类树状图上被划为另一大类 ,而且遗传距离与其它地理生态型也相差很远 .西藏高地核桃是作为一个地理生态型 ,还是作为一个独立的植物种有待进一步深入研究

Abstract:Eighty 10?bp random primers were used to amplify RAPD fragments for the 20 samples of 4 different walnut ecological types from Xinjiang, Huabei (North China) and Qinba(Southwest China) mountainous regions and Tibet. Among these primers, only 24 gave highly reproductive amplification and produced 227 RAPD fragments, 168 (74%) of which showed polymorphic with the size ranging from 220?bp to \{4 072\}?bp. On average 9.46 fragments per primer were amplified. The samples from Tibet exhibited many unique RAPD fragments. The genetic distances were estimated from these RAPD data. UPGMA method was employed in the cluster analysis to obtain the dendrogram. The latter indicates that Xinjiang walnut is a separate group of \%J. regia\%; Huabei walnut and Qinba walnut are two different subtypes in the same group. Not only is Tibet walnut distinguished from other groups, but also its genetic distance is significantly different from that of other types. However, Tibet walnut is distinguished as a group or an independent species still needs to be studied.
