京西葛组织培养及快速繁殖研究

摘    要:通过京西葛茎段的离体培养和快速繁殖实验,探讨了京西葛不定芽的诱导、分化、增殖及生根的最佳培养条件。结果表明:以优良京西葛的具腋芽茎段为外植体,经2.2%NaClO灭菌25~30min后成活率可达50%;适于京西葛不定芽诱导的培养基为MS+NAA0.1mg/L+6-BA0.5mg/L,诱导分化率为88.89%;适于不定芽分化及继代培养的培养基为MS+NAA0.15mg/L+6-BA0.3mg/L+2,4-D0.2mg/L,继代芽分化率为83.33%,继代倍数达6~7倍;适于生根的培养基为1/2MS+NAA0.5mg/L,生根率达100%。本研究最终建立了京西葛的组织培养体系并初步建立起一套较为完善的快速繁殖技术体系。

Abstract:The optimal tissue culture conditions for induction,differentiation,proliferation and rooting of adventitious buds of Pueraria lobata Ohwi were studied through in vitro culture and rapid propagation technologies.The results showed that using stem segments with axillary buds from elite parent trees of P.lobata as explants,the survival rate could reach 50% after 25-30 min of sterilization with 2.2% NaClO.The most optimal medium for induction of adventitious buds was MS + NAA 0.1 mg/L + 6-BA 0.5 mg/L,and the i...
