白、青杨派间杂交幼胚培养及杂种子代的分子鉴定

摘    要:利用切枝水培和组织培养技术,对白杨派的毛新杨和青杨派的不同种源小叶杨进行了派间杂交和杂交子代的幼胚培养研究。结果显示:以不同种源的小叶杨为父本和授粉后幼胚培养的时间对幼胚萌发率影响较大。其中毛新杨×小叶杨(内蒙古通辽)的幼胚培养效果最好,最适培养基为1/2MS+6BA 1.0 mg/L+NAA 1.0 mg/L;且随着授粉后天数的增加,幼胚培养的萌发率也相应增加。在此基础上,选用SSR标记技术对杂交子代及其亲本进行了鉴定分析。共采用120对杨树特异的SSR引物对杂交亲本毛新杨和小叶杨进行了多态性筛选,得到了25对在双亲间显示多态性的引物。对其中的两对进行了杂交子代的SSR扩增分析,结果表明,杂交子代均显示出双亲间的杂种类型谱带,可以在分子水平上证明白、 青杨派间的杂交可配性。

Abstract:Inter sectional hybridization between (P. tomentosa ×P. bolleana) and P. simonii and the immature ovule culture of the offspring were performed in this paper by the controlled hybridization and in vitro organ culture technology. The results indicate that different provenances of P. simonii as paternal parents and culture time of immature ovule after pollination had great influences on the germination rate of immature ovule. The highest germination rate of immature ovule was the crossing combination of (P. tomentosa ×P. bolleana) × P. simonii clone from Tongliao, Inner Mongolia Autonomous Region and the optimal culture medium was 1/2MS+6BA 1.0 mg/L+NAA 1.0mg/L. Meanwhile as the days increased after pollination, germination rate of immature ovule increased too. Simple sequence repeat (SSR) technique was used to identify the progeny of (P. tomentosa ×P. bolleana) and P. simonii. 120 primer pairs of Populus specific SSR were been screened to detect the differences between both parents, of which 25 primer pairs produced the polymorphic loci. Two of 25 were used to detect the offspring of (P. tomentosa ×P. bolleana) and P. simonii. The results show that all hybrids have the heterozygous bands caused by the recombination between the both parents. The molecular evidence indicates the reproductive fertility of inter sectional hybridization between (P. tomentosa ×P. bolleana) and P. simonii.
