胡杨叶片不定芽再生体系的研究

摘    要:该文以胡杨试管苗叶片为材料 ,进行了不同培养基及不同影响因子对胡杨叶片不定芽再生能力影响的试验 .结果表明 :胡杨叶片再生的最适培养基是MS +BA 0 .5mg L +NAA 0 1~ 0 2mg L +腺嘌呤 4 0mg L +水解乳蛋白 5 0 0mg L +白砂糖 2 5g L +琼脂 5g L ,再生频率达 10 0 % ;叶片培养的最佳取材位置是自茎尖展叶始第 1~ 3片叶 ;叶片正面向上或正面向下接种培养 ,对其形成不定芽无显著影响 ;由丛生芽和愈伤组织两个途径得到的无菌苗上的叶片在再生能力方面没有明显差异 .

Abstract:Adventitious bud regeneration from the leaves of Populus euphratica in vitro with different media and factors of tissue culture was studied. The results showed that the optimum medium for adventitious bud regeneration was MS+BA 0.5 mg/L+NAA 0.1~0.2 mg/L+Adenine 40 mg/L+Hydrolyzate lactalbumin 500 mg/L+sugar 25 g/L+agar 5 g/L, and the frequency of adventitious bud regeneration could reach 100%. The best leaves for adventitious bud regeneration are the first three top leaves of a shoot. There weren't significant effects on adventitious bud regeneration from the leaves which were cultured on medium with the adaxial side upward(abaxial side of leaf explant contacted with medium) or with the adaxial side downward (adaxial side of leaf explant contacted with medium). There isn't much difference in adventitious bud regeneration from leaves of proliferation of bud clusters or leaves of callus.
