农杆菌介导抗草甘膦基因(EPSPS)的玉米转化及相关因子的影响研究

摘    要:对影响农杆菌转化效率的一些因素进行了研究优化,结果表明,诱导培养基中添加200 mg/L的头孢霉素既能抑制农杆菌的生长又不影响幼胚的愈伤诱导;诱导和筛选培养基中添加5 mg/L硝酸银能有效抑制玉米愈伤组织褐变,显著改善愈伤组织及胚性愈伤组织的诱导率;分化培养基中添加2μmol/L的嘧啶醇能延缓试管苗的生长速度,使苗更健壮。利用优化的根癌农杆菌介导转化体系将抗草甘膦基因(EPSPS)转入玉米杂交材料HiⅡ中,获得了PCR阳性植株。

Abstract:Several factors influencing transformation efficiency were studied and optimized.It showed that,adding 200 mg/L Cofotaxime could inhibit the growth of Agrobacterium but not influence the callus induction;adding 5 mg/L AgNO_3 could inhibit maize callus browning,the regeneratet maize plantlets grew more slowly and robuster on the medium with 2μmol/L ancymidol.The glyphosate-resistant gene (EPSPS) was introduced into Hi II,a maize hybrid,by Agrobacterium tumefaciens method.The PCR positive plants were obtained.
