亚比棉×陆地棉杂种F_1遗传分析

摘    要:为了验证亚比棉×陆地棉杂种F1的真实性,从细胞学和RAPD技术方面对亚比棉×陆地棉杂种F1及其亲本进行了研究。细胞学研究表明,三种杂种(亚洲棉×比克氏棉)×陆地棉的核型公式为2n=4x=52=2M+44m(2SAT)+6sm(2SAT),相对长度变异范围为6.12%~2.56%,最长染色体与最短染色体之比为2.39∶1,臂比值在1.00~1.82之间,平均臂比为1.35,核型类别为1B。RAPD分析表明,亚比棉×陆地棉杂种F1共扩增出61条DNA带,其中具亚洲棉特异带的占总带数的3.3%,来自比克氏棉的带占9.8%,具陆地棉特异带的占13.1%,来自亚比棉的特异带占9.8%,杂种的特异带占14.8%。细胞学研究和RAPD分析的结果吻合,从而断定亚比棉×陆地棉杂种F1为真正的杂交种。

Abstract:To veritify the truth of hybrid,(G arboreum×G bickii)×G hirsutum and its parents were studied using cytology and RAPD technology.The result of cytology showed that the karyotype foumula of tri-hybrid(G arboreum×G bickii)×G hirsutum was 2n=4x=52=2M+44m(2SAT)+6sm(2SAT).The range of relative length was 6.12~2.56 %.The ratio is 2.39∶1,the range of arm ratios was 1.00~1.82 between the longest and the shortest chromosome with a mean arm ratios of 1.35.The type of karyotype is 1B.The analysis of RAPD showed: A total of sixty-one DNA bands were amplified in(G arboreum×G bickii)×G hirsutum,3.3%DNA bands came fromthe special bands of G arboreum,9.8%was fromthe special bands of G.bickii.13.1%DNA bands came from the special bands of G hirsutum,9.8%came from the special bands of(G arboreum×G bickii).14.8%DNA came from the special bands of tri-hybrid.The results of cytology and molecular marker showed that the tri-species of(G arboreum×G bickii)×G hirstum was true tri-specific hybrid.
